目的:优选茵陈四苓颗粒的乙醇提取工艺。方法:以栀子苷提取量为指标,通过单因素试验比较乙醇回流法、乙醇超声法和乙醇渗漉法,确定茵陈四苓颗粒中部分药材的提取方法;通过正交试验考察乙醇体积分数、浸泡时间、加醇量、渗漉流速对提取工艺的影响。采用HPLC测定栀子苷含量,色谱条件为UltimateTMC18色谱柱(4.6 mm×250 mm,5μm),流动相乙腈-水(15∶85),流速1 mL·min-1,柱温40℃,检测波长238 nm,进样量10μL。结果:采用乙醇渗漉法提取,最佳提取工艺条件为加10倍量70%乙醇浸泡12 h,渗漉流速5 mL·min-1,栀子苷提取量达42.524 4 mg·g-1。结论:该工艺客观可行、稳定合理,可为茵陈四苓颗粒的工业生产提供理论依据。
Objective: To optimize the ethanol extraction process of Yinchen Siling granule. Methods: The extraction of geniposide was used as an index to determine the extraction method of some herbs in Yinchen Siling Granules by ethanol refluxing, ethanol ultrasonic and ethanol percolation by single factor test. The orthogonal test was used to investigate the ethanol volume fraction, Time, add alcohol, percolation flow rate on the extraction process. The content of geniposide was determined by HPLC. The chromatographic conditions were: UltimateTMC18 column (4.6 mm × 250 mm, 5 μm), mobile phase acetonitrile-water (15:85), flow rate 1 mL · min- 238 nm, injection volume 10μL. Results: The optimum extraction conditions were as follows: ethanol percolation method, soaking for 10 h with 10 times the amount of 70% ethanol for 12 h, the percolation flow rate was 5 mL · min-1 and the extraction yield of geniposide was 42.524 4 mg · g-1. Conclusion: The process is objective and feasible, stable and reasonable, which can provide theoretical basis for the industrial production of Yinchen Siling granules.
