目的 研究B7瘤苗和负载了肿瘤细胞膜抗原的DC瘤苗在小鼠L6 15白血病模型中诱导抗白血病免疫的作用。方法 通过观察小鼠的存活率和生存时间 ,研究两种瘤苗在动物体内诱导免疫预防保护的能力 ;通过体外细胞毒实验和细胞增殖实验 ,检测两种瘤苗在体外诱导的T细胞特异性杀伤活性及对特异性抗原刺激的增殖活性。结果 在体内 ,两种瘤苗免疫的小鼠对随后活瘤细胞的攻击均有保护作用 ,DC瘤苗效果更佳 ;同时还发现输注免疫保护鼠的DC对荷瘤小鼠有明显的治疗效果。在体外 ,两种瘤苗均可诱导出对肿瘤细胞的特异性杀伤活性和对抗原的特异性增殖活性 ,DC瘤苗的活性更显著。结论 DC瘤苗以其安全、有效、易行的特点 ,将会在肿瘤免疫治疗中具有良好的应用前景。
Objective To study the anti-leukemic immunity induced by B7 tumor vaccine and DC vaccine loaded with tumor cell membrane antigen in mouse L6 15 leukemia model. Methods By observing the survival rate and survival time of mice, we studied the ability of the two kinds of tumor vaccine to induce immune protection and protection in animals. The in vitro cytotoxicity and T cell proliferation Killing activity and proliferating activity stimulated by specific antigen. Results In vivo, the mice immunized with the two kinds of vaccines had the protective effect on the subsequent live tumor cells, and the DC vaccine had a better effect. At the same time, it was found that DCs infused immunoprotective mice showed obvious treatment on tumor-bearing mice effect. In vitro, both tumor vaccine can induce specific cytotoxic activity against tumor cells and antigen-specific proliferative activity, DC tumor vaccine activity more pronounced. Conclusion DC vaccine is safe, effective and easy to use and will have a good prospect in tumor immunotherapy.
