目的 了解不同 +GZ作用下大鼠脑细胞凋亡的发生和凋亡相关基因 bcl- 2、p5 3基因表达的变化 ,探讨细胞凋亡在反复高 +GZ暴露所致脑损伤中的病理作用。 方法 48只 SD大鼠随机分成对照组、+7GZ暴露组、+10 GZ暴露组和 +14GZ暴露组 ,各组分别于暴露后 30 min、6 h、2 4h和 48h处死大鼠取脑。用原位末端标记法 ( TUNEL )检测大鼠脑细胞凋亡及用半定量聚合酶链反应检测凋亡相关基因 bcl- 2和 p5 3 m RNA表达的变化。 结果 +10 GZ暴露组和 +14GZ暴露组在 6 h和 2 4h时在大脑皮层、海马 CA1区和纹状体可见部分神经元细胞发生凋亡和 bcl- 2、p5 3 m RNA表达变化。 结论 反复高 +GZ暴露可引起大鼠脑内部分神经细胞凋亡及 bcl- 2和 p5 3的表达变化 ,细胞凋亡可能是反复高 +GZ暴露致脑损伤的机理之一。
Objective To investigate the changes of apoptosis in rat brain and the expressions of bcl-2 and p53 genes under different + GZ treatment and to explore the pathological role of apoptosis in brain injury induced by repeated high + GZ exposure. Methods 48 SD rats were randomly divided into control group, + 7GZ exposure group, +10 GZ exposure group and +14 GZ exposure group. Rats in each group were sacrificed at 30 min, 6 h, 24 h and 48 h after exposure. The apoptotic brain cells were detected by TUNEL and the expressions of bcl-2 and p5 3 mRNA were detected by semi-quantitative polymerase chain reaction. Results Apoptosis and expression of bcl-2 and p5 3 mRNA in some neurons were observed in the hippocampus CA1 and striatum of +10 GZ exposure group and +14 GZ exposure group at 6 h and 24 h. Conclusion Repeated high + GZ exposure may induce some neuronal apoptosis and changes of bcl-2 and p5 3 expression in rat brain. Apoptosis may be one of the mechanisms of repeated high + GZ exposure induced brain injury.
