目的:直接进样高效液相色谱法测定人血浆中的头孢唑林、头孢曲松、头孢唑肟的含量。方法:选用浸透限制固定相,紫外检测器,使三种头孢菌素分别在流动相pH5.75、5.25和4.35条件下与内标及血浆中的蛋白质得到良好的分离。结果:制作了三种头孢菌素各自的标准曲线,测定方法的变异系数在1.2%~7.1%,回收率近100%。实测了6例临床静脉输注头孢唑林、头孢曲松、头孢唑肟病人的血药浓度。结论:浸透限制固定相色谱法测定血浆中的头孢菌素,省去了前处理,快速简单,更适用于临床治疗药物监测。
OBJECTIVE: To determine the contents of cefazolin, ceftriaxone and ceftizoxime in human plasma by direct injection high performance liquid chromatography. Methods: The three stationary phases and the UV detector were used to separate the three cephalosporins from the internal standard and the plasma proteins under the conditions of mobile phase of pH5.75, 5.25 and 4.35, respectively. Results: Three standard curves of cephalosporins were prepared. The coefficient of variation (CV) was 1.2% -7.1% and the recovery was nearly 100%. 6 cases of clinical intravenous cefazolin, ceftriaxone, ceftizoxime patients blood concentration. Conclusion: The determination of cephalosporins in blood plasma by the limit-of-penetration fixed-phase chromatography eliminates the need for pre-treatment and is quick and easy. It is more suitable for the monitoring of clinical therapeutic drugs.
