[目的]探讨三氧化二砷(As2O3)对人肺腺癌A549细胞的放射增敏作用。[方法]MTT法确定As2O3对A549细胞的半数致死浓度(LD50),采用集落形成法观察20%LD50的As2O3对A549细胞的放射增敏作用。[结果]细胞生长抑制随As2O3浓度的增加而增强。As2O3+照射组的细胞存活率低于单纯照射组,D0值小于单纯照射组(1.46Gyvs2.03Gy),Dq值也小于单纯照射组(0.49Gyvs1.35Gy),存活曲线肩区(Dq)变小,放射增敏比(SER)为1.39。[结论]As2O3对肺腺癌A549细胞有明显的放射增敏作用,抑制A549细胞的修复能力使照射后细胞存活分数降低是其放射增敏的可能机制。
[Objective] To investigate the radiosensitization effect of arsenic trioxide (As2O3) on human lung adenocarcinoma A549 cells. [Method] The half-lethal concentration (LD50) of As2O3 on A549 cells was determined by MTT assay. The radiosensitization of A549 cells treated with 20% LD50 As2O3 was observed by colony formation assay. [Result] The cell growth inhibition increased with the increase of As2O3 concentration. The cell viability of As2O3 + irradiation group was lower than that of the irradiation alone group. The D0 value was lower than that of the pure irradiation group (1.46Gyvs2.03Gy), the Dq value was also lower than that of the pure irradiation group (0.49Gyvs1.35Gy), the survival curve shoulder area (Dq) The radiosensitization ratio (SER) was 1.39. [Conclusion] As2O3 has a significant radiosensitization effect on lung adenocarcinoma A549 cells. Decreasing the survival rate of A549 cells after radiotherapy is a potential mechanism of radiosensitization.
