用基因交换的方法将烟草花叶病毒普通株(中国分离物,TMV-Cv和番茄花叶病毒弱毒疫苗N14基因组的运动蛋白(MP)、外壳蛋白(CP)基因编码区和3'非编码区嵌合于N14和TMV-CV 5'非编码区和复制酶基因的下游,构建重组病毒克隆pTN和pNT,并进行体外转录和烟草侵染试验结果 pTN和 pNT体外转录物对烟草均具侵染性:前者感染枯斑三生烟( Nicotiana tobacum cv.Samsun NN),等量感染的枯斑形成数比 PTMV-Cv少;感染普通烟( Nicotiana tobacum cv. Huangm-iaoyu,黄苗榆品种),出现典型系统症状,但叶片畸形率较低.后者感染枯斑三生烟,等量感染的枯斑形成数比pN14多;感染普通烟仍不出现系统症状.结果表明:重组病毒TN和NT仍能在试验用烟草中增殖,前者在普通烟上基本表现为TMV-Cv的强病毒性状,后者在普通烟上表现为N14的弱病毒性状; TMV-CV和 N14的复制酶与 MP, CP和 3'非编码区之间为非严紧型连接,两者在类似的各亚基因组之间可进行功能互补.初步推测,N14复制酶编码基因的突变区对N14的致弱起主要作用;其运动蛋白编码基因的突变区对被侵染烟草
