目的 :观察中药半边旗二萜类化合物 5F对体外培养的人翼状胬肉成纤维细胞 (HPF)增殖周期和Ki 6 7表达的影响。方法 :用流式细胞仪 (FCM)分析不同浓度 5F对HPF周期分布、免疫细胞化学法 (ICH)检测 5F对增殖指数Ki 6 7抗原的影响。结果 :用药后HPF停滞于G1期 ,S、G2 /M期细胞减少 ;Ki 6 7阳性表达率对照组为 (2 6 16± 1 2 6 ) % ,5F浓度 8μg/ml时下降 ,在 12 8μg/ml时降至 (4 10± 0 6 3) %。各用药组与对照组比较均有显著性差异 (P <0 0 1)。各用药组间两两比较有显著性差异 (P <0 0 1)。结论 :5F对HPF增殖的抑制作用与细胞周期、Ki 6 7阳性表达降低有关
Objective: To observe the effect of diterpene 5F on the proliferation and Ki67 expression of human pterygium fibroblast (HPF) in vitro. Methods: The distribution of HPF at different concentrations of 5F was analyzed by flow cytometry (FCM), and the effect of 5F on the proliferation index Ki 6 7 antigen was detected by immunocytochemistry (ICH). Results: After treatment, HPF was arrested in G1 phase and decreased in S, G2 / M phase. The positive rate of Ki67 was (2 16 16 ± 1 26)% in control group and decreased in 5 8F 8μg / / ml to (4 10 ± 0 6 3)%. Each drug group and control group were significantly different (P <0.01). There was a significant difference between every two groups (P <0.01). CONCLUSION: The inhibitory effect of 5F on HPF proliferation is related to the decrease of cell cycle and Ki 6 7 expression
