目的:探讨丙酮酸乙酯对急性胰腺炎(AP)组织细胞线粒体功能的影响。方法:按随机数字表法将SD大鼠分为3组:A组(AP模型+REPS复苏组,20只)、B组(AP模型+RLS复苏组,20只)和C组(假手术组,20只)。采用向胆胰管内加压注入4%牛磺胆酸钠溶液的方法制作AP模型,术后24 h获取胰腺线粒体标本。检测Na~+K~+-ATP酶、Ca~(2+)Mg2~(2+)-ATP酶、总ATP酶活性、线粒体微黏度、线粒体膜脂质堆积密度。结果:3组大鼠胰腺线粒体Na~+K~+-ATP酶、Ca~(2+)Mg2~(2+)-ATP酶及总ATP酶活性、线粒体微黏度、线粒体膜脂质堆积密度水平比较,差异有统计学意义(P<0.05),其中A组与c组比较,差异无统计学意义(P>0.05),而B组与C组比较,差异有统计学意义(P<0.05),A组较B组效果显著(P<0.05)。结论:丙酮酸乙酯可以有效改善AP模型胰腺线粒体功能、缓解线粒体损伤、保护线粒体膜正常结构。
Objective: To investigate the effect of ethyl pyruvate on the mitochondrial function of acute pancreatitis (AP) cells. Methods: The SD rats were divided into three groups according to random number table: group A (AP model + REPS recovery group, 20 rats), group B (AP model + RLS recovery group, 20 rats) and group C (sham operation group , 20). AP model was made by injecting 4% sodium taurocholate into biliary and pancreatic ducts, and pancreatic mitochondrial specimens were obtained 24 hours after operation. The activities of Na ~ + K ~ + -ATPase, Ca ~ (2+) Mg2 ~ (2 +) - ATPase, total ATPase activity, mitochondrial microviscosity and mitochondrial membrane lipid bulk density were measured. Results: The activities of mitochondrial Na ~ + K ~ + -ATPase, Ca ~ (2+) Mg2 ~ (2 +) - ATPase and total ATPase activity, mitochondrial microviscosity, mitochondrial membrane lipid accumulation density (P <0.05). There was no significant difference between group A and group c (P> 0.05), but the difference between group B and group C was statistically significant (P <0.05) , A group than B group significant effect (P <0.05). Conclusion: Ethyl pyruvate can effectively improve mitochondrial function, alleviate mitochondrial damage and protect the normal structure of mitochondrial membrane in AP model.
