目的:研究腺病毒介导的多基因(GM-CSF、B7-1、p53、IL-2)在胆管瘤细胞系QBC939中的表达及对其生长的影响。方法:构建同时含GM-CSF、B7-1、p53、IL-24种目的基因的重组腺病毒载体Admultigenes,应用流式细胞仪、ELISA、免疫织化等方法,分析该重组腺病毒体包含的目的基因在靶细胞中的表达,并绘制细胞的生长曲线。结果:腺病毒介导的多基因在胆管癌细胞系QBC939中高效表达(IL-2不表达),并能抑制胆管瘤细胞的生长。结论:腺病毒介导的多种基因能在胆管瘤细胞QBC939中表达.并抑制肿瘤细胞的生长。
Objective: To investigate the expression of adenovirus-mediated polygenes (GM-CSF, B7-1, p53, IL-2) in the cholangiocarcinoma cell line QBC939 and its effect on growth. METHODS: Adenogenes, a recombinant adenoviral vector containing both GM-CSF, B7-1, p53, and IL-24 genes, was constructed and analyzed by flow cytometry, ELISA, and immunohistochemistry. The expression of the target gene in the target cell, and draw the cell growth curve. RESULTS: Adenovirus-mediated polygenes were highly expressed in the cholangiocarcinoma cell line QBC939 (IL-2 was not expressed) and inhibited the growth of cholangiocarcinoma cells. Conclusion: Adenovirus-mediated multiple genes can be expressed in cholangiocarcinoma cells QBC939. And inhibit the growth of tumor cells.
