目的:观察蒡芎疏风颗粒(Bxsfkl)对实验性偏头痛的影响。方法:将小鼠分为空白、模型、实验药物3个剂量和中、西药对照组,以Bxsfkl低、中、高(9.75,19.5,39 g.kg-1.d-1)ig给药干预5 d,检测各组小鼠的物理和化学痛阈值。利用硝酸甘油制造偏头痛模型,观察治疗后各组大鼠行为变化和血浆5-羟色胺(5-HT)测值,考察Bxsfkl对实验性偏头痛的效应。结果:在小鼠甩尾实验中,Bxsfkl高剂量组提高小鼠痛阈值,在30,60,90,120 min分别与空白组比较,有统计学意义(P<0.05或P<0.01)。在小鼠醋酸扭体实验中,Bxsfkl中、高剂量组减少小鼠扭体次数,与空白组比较有统计学意义。在硝酸甘油所致偏头痛模型实验中,Bxsfkl低、中、高剂量组大鼠耳红消失时间与模型组比较均有显著差异;大鼠挠头次数在30~180 min与模型组比较,均能缓解挠头次数,高剂量组有统计学意义;大鼠爬笼次数在30~180 min与模型组比较,其中高剂量组有统计学意义。大鼠血浆5-HT含量模型组为(43.99±5.49)ng.g-1较正常对照组(82.62±11.18)ng.g-1显著降低(P<0.01);与模型组比较,各治疗组血浆5-HT含量增加,且均有统计学意义。结论:Bxsfkl能明显提高小鼠的痛阈,显著缓解模型大鼠因头痛所致耳红、挠头、爬笼等现象,具有增加血浆中5-HT含量的作用。
Objective: To observe the effect of BXsfkl on experimental migraine. Methods: The mice were divided into three groups: blank, model, experimental drug and western medicine control group, with Bxsfkl low, medium and high (9.75, 19.5, 39 g.kg-1.d-1) After 5 days, the physical and chemical pain thresholds in each group were detected. Migraine model was made by using nitroglycerin, and behavioral changes and 5-hydroxytryptamine (5-HT) were measured in all groups to investigate the effect of Bxsfkl on experimental migraine. Results: In mice tail flick test, Bxsfkl high dose group increased the pain threshold in mice, and there was statistical significance (P <0.05 or P <0.01) at 30, 60, 90 and 120 minutes respectively compared with the blank group. In mice acetic acid writhing experiments, Bxsfkl, high dose group to reduce the number of writhing in mice, compared with the blank group was statistically significant. In nitroglycerin-induced migraine model experiments, the disappearance time of the ear red of Bxsfkl low, medium and high dose groups were significantly different compared with the model group; the number of scratching of the rats in 30 ~ 180 min compared with the model group Relieve the number of scratching the head, high-dose group was statistically significant; crawling cage number in 30 ~ 180 min compared with the model group, in which the high-dose group was statistically significant. The level of 5-HT in model group was significantly lower than that in normal control group (43.99 ± 5.49) ng.g-1 (82.62 ± 11.18 ng.g-1) (P <0.01). Compared with model group, Plasma 5-HT content increased, and both were statistically significant. CONCLUSION: Bxsfkl can significantly increase the pain threshold in mice and significantly alleviate the effects of ear-redness, scratching-head and climbing on the cage caused by headache, and increase the 5-HT content in plasma.
