绿豆种子萌发率严重影响和制约其田间产量,研究利用与绿豆种子萌发相关的基因资源具有十分重要的意义.本研究拟通过查询绿豆转录组数据库,分离包含完整ORF的生长素响应因子(ARF)基因家族成员,对比分析该家族成员的蛋白保守结构域,基于氨基酸序列同源性构建系统进化树.结果表明:分离到22个包含完整ORF的ARF基因,其核苷酸序列长度范围为905~3 624 bp,其编码氨基酸序列长度的范围为237~761 aa.其中仅有4个ARF基因包含保守的B3 DNA结合结构域、Auxin响应因子和AUX/IAA家族蛋白结构域,其他基因存在1~2个保守结构域的缺失.系统进化树结果显示,绿豆22个ARF基因与拟南芥12个ARF基因共划分为3大类,9个亚类.这些结果为进一步揭示绿豆ARF基因功能多样性,同时为将来遗传工程改良绿豆品种提供理论参考.“,”The yield of mung bean in the field was easily affected and restricted by seeds germination rates.It was very meaningful to research and utilize these genes resources related with seeds germination in mung bean.In this study,ARF gene family was investigated from the annotation of leaves transcriptome data in mung bean,and isolated to acquire genes including complete ORF region.Comparing among these genes with homologous proteins in A rabidopsis thaliana,their conserved domain was analyzed,and the phylogenetic tree was constructed.The results showed that 22 ARF genes with complete ORF region were identified,and their nucleotide sequence length was ranged from 905 to 3 624 bp,which encoded amino acid from 237 to 761 amino acid residues.Only 4 ARF genes contained conserved B3 DNA binding domain,Auxin response factor,and Aux/IAA family,the others had one or two conserved domain absence in their protein.The results ofphylogenetic tree indicated that 22 ARF genes in mung bean and 12 ARF genes in Arabidopsis thaliana were classified into 3 group and 9 subgroups.These results suggested that these ARF genes have functional diversity in mung bean.It would be laid the foundation for genetic modified and improved germplasm resources of mung bean in the future.
