目的建立采用快速溶剂萃取技术处理固体样品,应用高效液相色谱—紫外检测器同时测定食品中酸性橙Ⅱ染料和柠檬黄、苋菜红、胭脂红、日落黄、亮蓝5种合成色素的方法。方法液体样品经过稀释、过膜等简单处理后直接进样,固体样品经过快速溶剂萃取技术提取浓缩后进样,色谱柱采用YWG C18 250×4.6mm,10μm,流动相为甲醇∶0.02mol/L乙酸铵进行梯度洗脱,流速为0.6~1.0ml/min,检测波长为484和600nm进行检测。结果液体样品平均回收率为95.3%~99.0%,RSD为0.3%~3.4%;固体样品平均回收率为84.3%~95.2%,RSD为0.7%~4.6%。6种物质的线性范围为2.5~25μg/ml,相关系数为0.99978~0.99998,检出限为柠檬黄0.15μg/ml,苋菜红0.20μg/ml,胭脂红0.18μg/ml,日落黄0.20μg/ml,亮蓝0.18μg/ml,酸性橙Ⅱ0.20μg/ml。结论建立的方法用于同时测定6种物质,分离度好,回收率高,适合于日常检测工作。
OBJECTIVE To establish a method for the simultaneous determination of 5 kinds of synthetic pigments including Acid Orange II dye, Lemon Yellow, Amaranth Carmine, Carmine, Sunset Yellow and Bright Blue by using the rapid solvent extraction technology and high performance liquid chromatography - ultraviolet detector. Methods The liquid sample was diluted and passed through the membrane. The solid sample was directly extracted by rapid solvent extraction and concentrated. The sample was packed with YWG C18 250 × 4.6 mm, 10 μm and the mobile phase consisted of methanol: 0.02 mol / L Ammonium acetate gradient elution, flow rate of 0.6 ~ 1.0ml / min, the detection wavelength of 484 and 600nm were detected. Results The average recoveries of liquid samples ranged from 95.3% to 99.0% with RSDs of 0.3% to 3.4%. The average recoveries of solid samples ranged from 84.3% to 95.2% with RSDs ranging from 0.7% to 4.6%. The linear range of the six substances was 2.5 ~ 25μg / ml, the correlation coefficient was 0.99978 ~ 0.99998, the detection limits were 0.15μg / ml of tainted yellow, 0.20μg / ml of amaranth, 0.18μg / ml of carmine and 0.20μg / ml, Brilliant Blue 0.18μg / ml, Acid Orange II 0.20μg / ml. Conclusion The established method is suitable for the simultaneous determination of six substances with good resolution and high recovery rate and is suitable for routine testing.
