目的:选用壳聚糖澄清剂替代传统醇沉方法对板蓝根水提液进行纯化,改进板蓝根的纯化工艺条件,节约工艺成本。方法:通过检测纯化后的药液的透光率、固形物保留率及腺苷含量来考察絮凝温度、药液pH、壳聚糖加入量三个因素对絮凝效果的影响;并进行絮凝温度、药液pH、壳聚糖加入量和絮凝时间四因素三水平正交试验。结果:在絮凝温度60℃、药液pH为4、壳聚糖加入量为2 ml·g~(-1)、絮凝时间12 h条件下,壳聚糖絮凝剂对板蓝根水提液有良好的絮凝作用。结论:壳聚糖絮凝工艺可替代传统醇沉工艺用于板蓝根的纯化,可节省生产成本。
OBJECTIVE: To purify the aqueous extract of Radix Isatidis by using chitosan clarifier instead of the traditional alcohol precipitation method to improve the purification process conditions of Radix isatidis and save the process cost. Methods: The effects of flocculation temperature, liquor pH and the dosage of chitosan on the flocculation efficiency were investigated by measuring the light transmittance, retention rate and adenosine content of the purified liquid. The flocculation temperature, Liquid pH, chitosan dosage and flocculation time of four factors and three levels of orthogonal test. Results: Under the condition of the flocculation temperature of 60 ℃, pH value of solution 4, the amount of chitosan added 2 ml · g -1, flocculation time 12 h, the chitosan flocculant had a good Flocculation. Conclusion: Chitosan flocculation process can replace the traditional alcohol precipitation process for the purification of Banlangen, saving production costs.
