IL-2、IFN-α基因转染的人肾癌细胞体外生长及体内致瘤性下降,本文进一步研究了IL-2和/或IFN-α基因转染对人肾癌R11细胞浸润性和转移性的影响.恶性肿瘤的转移与浸润与肿瘤细胞表面的某些粘附分子(如ICAM-1,CD54)与细胞外基质成分(如Fibornectin,Colla-gen,Laminin,Vitronectin)相互作用有关.采用FACS分析了IL-2和IFN-α基因转染的R11细胞,包括R11-IL-2(H)(高分泌IL2细胞株,2000U/1O~6天)、R11-IL-2(L)(低分泌IL-2细胞株,25U/10~6/天)、R11-IFN-α(H)(4000U/10~6/天)、R11-IFN-α(L)96U/10~6天)、R11-IL-2+IFN-α(IL-295U,IFN-α1270U/10~6/天)以及转染对照质粒的R11细胞表面的ICAM-1表达水平和CD44的表达水平,结果发现:除R11-IFN-α(H)细胞外(可能由于高水平的IFN-α抑制了细胞的蛋白质合成),其余细胞因子转染后的R11细胞表面ICAM-1表达均明显高于转染对照质粒的R11细胞;所有细胞因子基因转染的R11细胞表达CD44的表达量均减少.向预先覆盖有细胞外基质成分的96孔板中加入~3H-TdR标记的R-11细胞检测细胞与基质的粘附程度,结果显示,所有细胞因子转染后的R11细胞与细胞外基质的粘附均明显减弱.另外,IL-2和/或IFN-α细胞因子基因转染的同种肿瘤细胞间聚集明显降低.上述结果均与这些细胞因子基因转染后的
