目的:研究黑三棱花粉活力与柱头可授性,为杂交育种工作提供理论依据。方法:采用琼脂培养基萌发法测定花粉活力,研究不同培养基、采集时间、贮藏条件对花粉活力的影响;用联苯胺-过氧化氢法测定柱头可授性。结果:最佳的花粉培养基为:10%蔗糖+1%琼脂+0.01%MgSO4·7H2O+0.01%Ca(NO3)2·3H2O;开花第3天柱头可授性最强;低温干燥能延长花粉的贮藏时间,-70℃干燥条件下花粉可贮藏50 d。结论:黑三棱人工授粉的最佳时期为开花后2~3 d。
Objective: To study the pollen viability and stigma receptivity of Black Triangulation and provide a theoretical basis for the work of hybrid breeding. Methods: Pollen viability was determined by agar medium germination method. The effects of different media, collection time and storage conditions on pollen viability were studied. The stigma receptivity was determined by benzidine - hydrogen peroxide method. Results: The best pollen medium was: 10% sucrose + 1% agar + 0.01% MgSO4 · 7H2O + 0.01% Ca (NO3) 2 · 3H2O; stigma receptivity was the highest on the third day of flowering; The storage time, -70 ℃ dry conditions pollen can be stored for 50 d. Conclusion: The optimal period of Artificial pollination is 2 ~ 3 d after flowering.
