目的探讨依达拉奉对帕金森病(PD)小鼠黑质多巴胺神经元的保护作用。方法 30只健康雄性C57BL/6小鼠,随机分为正常对照组、模型组和治疗组。后两组给予腹腔注射百草枯和代森锰建立PD模型。建模成功后,治疗组给予依达拉奉腹腔注射2周,正常对照组给予同剂量生理盐水。观察3组小鼠行为学的变化,并用免疫组织化学法检测黑质TH、Caspase-3的表达。结果与治疗组比较,模型组小鼠爬杆时间明显延长,黑质TH阳性神经元数量显著降低,Caspase-3的表达显著增高。结论依达拉奉治疗可使PD小鼠运动能力增强,黑质TH阳性神经元数量增加,并降低Caspase-3的表达,有明显的神经保护作用。
Objective To investigate the protective effect of edaravone on substantia nigra dopamine neurons in Parkinson’s disease (PD) mice. Methods Thirty healthy male C57BL / 6 mice were randomly divided into normal control group, model group and treatment group. The latter two groups were given intraperitoneal injection of paraquat and dexmedetomidine PD model. After modeling, the treatment group was given edaravone intraperitoneal injection for 2 weeks, the normal control group given the same dose of saline. The behavioral changes of the three groups were observed, and the expression of TH and Caspase-3 in substantia nigra was detected by immunohistochemical method. Results Compared with the treatment group, the climbing time of mice in the model group was significantly prolonged, the number of TH substantia nigra neurons was significantly reduced, and the expression of Caspase-3 was significantly increased. Conclusion Edaravone can enhance the motor ability of PD mice, increase the number of TH positive substantia nigra neurons and decrease the expression of Caspase-3, which has a significant neuroprotective effect.
