自1971年Bouvier发现乙型肝炎病毒(HBV)表面抗原(HBsAg)两个独立的点突变决定簇d和y以来,许多学者对HBsAg亚型进行了深入的研究,先后发现了HBsAg多种亚型。1997年Lindh等用核酸测序法对166例HBV聚合酶链反应(PCR)阳性病人的S基因限制性片段长度多态性进行了分析,鉴别出6种基因型。由于PreS2区基因比S区基因更易突变.故作者选择PreS2区基因表达产物来制备单克隆抗体以研究HBV基因型。
Since Bouvier’s discovery of two independent point mutation determinants d and y of hepatitis B virus surface antigen (HBsAg) in 1971, many scholars have conducted an in-depth study of the HBsAg subtypes, and have discovered many subtypes of HBsAg . In 1997, Lindh et al. Used nucleic acid sequencing to analyze the restriction fragment length polymorphism of S gene in 166 patients with HBV polymerase chain reaction (PCR) positive, and identified 6 genotypes. Because the PreS2 gene is more susceptible to mutation than the S gene, the authors selected the PreS2 gene expression product to prepare a monoclonal antibody to study the HBV genotype.
