[目的]从糖尿病多因素致病角度出发,探讨情志因素对2型糖尿病发病的作用机制———气机失调证,以及具有理气降逆散结功效的复方中药心身5号方的调节作用。[方法]采用旋转、拥挤、限制3种情志刺激结合腹腔注射少量链脲佐菌素(STZ)的实验方法,制作了2型糖尿病倾向大鼠模型,实验动物分为正常组、应激组、STZ组、应激+STZ组和中药组。检测血浆去甲肾上腺素(NE)、肾上腺素(E)含量,同时观察具有理气降逆散结功效的复方中药心身5号方的调节作用。[结果]慢性情志应激可促使STZ鼠血浆NE、E含量升高,其中肾上腺素在应激+STZ组有极显著性差异(P<0.01),中药可降低血浆E含量。[结论]慢性情志刺激引起实验性2型糖尿病倾向大鼠血浆NE、E含量升高是糖尿病发病机制之一,同时具有理气降逆散结功效的复方中药心身5号方有明显调节作用。
[Objective] To explore the mechanism of affective factors on the pathogenesis of type 2 diabetes mellitus (DM) from the perspective of multifactorial diabetes mellitus (T2DM), and the regulatory effect of TCM Formula No. 5 . [Method] The type 2 diabetic predisposed rat model was made by rotating, crowding, limiting the 3 kinds of emotional stimuli and intraperitoneal injection of a small amount of streptozotocin (STZ). The experimental animals were divided into normal group, stress group , STZ group, stress + STZ group and TCM group. The levels of norepinephrine (NE) and epinephrine (E) in plasma were measured. Meanwhile, the regulation effect of compound Chinese medicine Xinshitang 5 with the effect of reducing blood pressure and descending nodules was observed. [Results] Chronic emotional stress could promote the plasma NE and E levels in STZ rats. There was a significant difference in epinephrine between stress + STZ group (P <0.01), and Chinese medicine could decrease plasma E content. [Conclusion] The increase of plasma NE and E levels induced by chronic emotional stimulation in experimental type 2 diabetic rats is one of the pathogenesis of diabetes mellitus. At the same time, the compound Chinese herbal medicine Xinshitang No.5 with the effect of reducing qi, reducing blood pressure and reducing diarrhea has obvious regulation.
