免疫应答期间脑和淋巴器官内儿茶酚胺代谢的动态变化

目的 :研究在免疫应答期间 ,脑和淋巴器官中儿茶酚胺代谢的动态变化 ,藉以了解免疫状态对中枢和外周儿茶酚胺能神经活动的影响。方法 :以绵羊红细胞作为抗原免疫 Sprague- Dawley大鼠 ,在免疫后的第 2、3、4、5、6、7d,应用高效液相色谱—电化学检测法测定大鼠下丘脑、海马、脑干和胸腺中去甲肾上腺素、肾上腺素、多巴胺和高香草酸的含量。结果 :1下丘脑去甲肾上腺素在免疫后第 4~ 7d都明显高于盐水对照组 ;海马内去甲肾上腺素只在免疫后第 4、5 d明显升高 ;胸腺中去甲肾上腺素在免疫后第 4、5 d明显降低。 2下丘脑肾上腺素在免疫后第 4~ 6 d显著增加 ;海马内肾上腺素在免疫后第 5、6 d明显增多 ;胸腺中肾上腺素明显增加发生在免疫后第 6、7d。 3下丘脑多巴胺在免疫后第 3、4d明显升高 ,而在第 5 d明显降低 ,高香草酸在免疫后第 2~ 5 d显著升高 ;海马内多巴胺在免疫后第 5~ 7d明显降低 ,而高香草酸在第 5~ 7d明显升高 ;胸腺中多巴胺只在免疫后第 6 d显著增加。4脑干中儿茶酚胺含量在免疫应答期间均无显著变化。结论 :免疫应答可能使下丘脑和海马中儿茶酚胺能神经的活动加强 ,对胸腺儿茶酚胺的影响可能依递质不同而作用不同 ,但对脑干中儿茶酚胺能神经的活动可能没有明显的影响 OBJECTIVE: To study the dynamic changes of catecholamine metabolism in brain and lymphoid organs during the immune response in order to understand the effects of immune status on central and peripheral catecholaminergic activity. Methods: Sprague-Dawley rats were immunized with sheep erythrocytes as antigens. Hypothalamus, hippocampus and brain were measured by high performance liquid chromatography-electrochemical detection at 2, 3, 4, 5, 6 and 7 days after immunization Norepinephrine, epinephrine, dopamine and homovanillic acid in the stem and thymus. Norepinephrine in the hypothalamus was significantly higher than saline control group from the 4th to the 7th day after immunization. The norepinephrine in the hippocampus was increased only on the 4th and 5th day after immunization. The level of norepinephrine in the thymus 4 d after immunization was significantly reduced. Hypothalamic epinephrine increased significantly from 4 to 6 days after immunization. Adrenaline in hippocampus significantly increased at 5 and 6 days after immunization. Adrenaline increased significantly in thymus at 6 and 7 days after immunization. The hypothalamus dopamine increased significantly on the 3rd and 4th day after immunization, but decreased significantly on the 5th day. The homovanillic acid increased significantly on the 2nd to 5th day after immunization. The dopamine in the hippocampus decreased significantly on the 5th and 7th day after immunization , While that of vanillic acid increased significantly from the 5th to 7th day. The dopamine in the thymus increased significantly only on the 6th day after immunization. No significant changes in brain stem catecholamine levels during the immune response. CONCLUSIONS: Immune response may enhance the activity of catecholaminergic neurons in the hypothalamus and hippocampus. The effect on catecholamines in the thymus may be different depending on the neurotransmitter but may not have a significant effect on the activity of catecholaminergic nerves in the brain stem
