转基因小麦B72-8-11b中外源品质基因1Dx5表达量是内源相应基因表达量的6倍。利用小麦转基因品系为父本,常规品种鄂麦12、鄂麦18和日喀则8号分别为母本,配置3个杂交组合。采用SDS-PAGE技术,检测各组合亲本、F1、F2代的HMW-GS组成,研究转基因小麦B72-8-11b中外源品质基因1Dx5表达的遗传。结果表明:外源1Dx5基因有功能拷贝整合在1个位点,如同内源品质基因,遵从孟德尔遗传模式。这对育种选择策略的制订具有指导意义,也表明了基因枪转化法能够使得外源基因有功能拷贝整合在1个位点并能稳定传递。
The expression level of foreign gene 1Dx5 in transgenic wheat B72-8-11b is 6 times that of the endogenous corresponding gene. The transgenic wheat lines were used as male parents. The conventional breeds Emai 12, Emai 18 and Shigatan 8 were female parents respectively, and three crosses were arranged. The composition of HMW-GS in parents, F1 and F2 of each combination was detected by SDS-PAGE, and the inheritance of 1Dx5 gene expression in transgenic wheat B72-8-11b was studied. The results showed that the functional copy of the exogenous 1Dx5 gene was integrated into one locus, which was similar to the endogenous quality gene and followed the Mendelian inheritance pattern. This is of guiding significance for the formulation of breeding selection strategies and also shows that the gene gun transformation method enables the functional copy of the foreign gene to be integrated at one locus and can be transmitted stably.
