目的 探讨正常造血细胞中血管内皮生长因子 (VEGF)基因的表达水平。方法 TRIzol一步法提取正常外周血和非恶性骨髓标本单个核细胞及正常血小板的总RNA ;以 β2 -微球蛋白基因为内对照 ,逆转录聚合酶链反应 (RT -PCR)半定量分析VEGF基因的表达。结果 2 2份外周血标本有 18份表达VEGF基因 ,表达率为81.8% ,其中 8份为中度表达 ;18份骨髓标本VEGF基因的表达率高达 94.4% (17/ 18) ,其中高度表达和中度表达分别为 5份与 6份 ;血小板有VEGF基因的低度表达。结论 正常造血细胞可表达VEGF基因 ,提示体内血管生成受到多种效应细胞的共同调节
Objective To investigate the expression of vascular endothelial growth factor (VEGF) gene in normal hematopoietic cells. Methods Total RNA was extracted from mononuclear cells and normal platelets of normal peripheral blood and non-malignant bone marrow samples by one-step TRIzol method. VEGF gene was semi-quantitatively analyzed by reverse transcription polymerase chain reaction (RT-PCR) with β2-microglobulin gene as control. expression. Results The VEGF gene was expressed in 18 out of 22 peripheral blood samples and the expression rate was 81.8%, of which 8 were moderately expressed. The expression rate of VEGF gene in 18 bone marrow specimens was as high as 94.4% (17/18) Moderate expression of 5 copies and 6 copies; Platelets have low expression of VEGF gene. Conclusion Normal hematopoietic cells can express VEGF gene, suggesting that angiogenesis in vivo is regulated by a variety of effector cells
