目的观察磷酸川芎嗪(LP)贴剂对大鼠急性心肌缺血的保护作用及凝血时间的影响。方法采用舌下静脉注射垂体后叶素(Pit)致大鼠急性心肌缺血模型,将大鼠随机分为模型组、赋形剂组、阳性组及LP贴剂高、中、低(4、2、1 cm2)剂量组等6组。观察各组大鼠注射Pit后不同时间点的Ⅱ导联心电图S-T段及T波的变化情况。采用毛细管法于眼眶取血,测定大鼠的凝血时间。观察LP贴剂3个剂量组与赋形剂组对大鼠凝血时间的影响。结果 LP贴剂高、中、低剂量组均能明显抑制注射Pit后15 s~5 min内大鼠心电图S-T段与T波的变化,均能明显延长大鼠的凝血时间。结论 LP贴剂对Pit致大鼠急性心肌缺血有明显的保护作用,具有抗凝血即抑制血小板聚集的作用。
Objective To observe the protective effect of ligustrazine phosphate (LP) on acute myocardial ischemia in rats and the effect of clotting time. Methods The rat model of acute myocardial ischemia induced by pituitary injection of pituitary was used. The rats were randomly divided into model group, vehicle group, positive group and LP patch high, medium and low (4, 2,1 cm2) dose group 6 groups. The changes of S-T segment and T wave of Ⅱ lead electrocardiogram were observed at different time points after injection of Pit in each group. Using capillary method in the orbital blood, measuring the clotting time of rats. To observe the effect of 3 patches and excipient of LP patch on clotting time of rats. Results LP patch of high, medium and low dose group can significantly inhibit the changes of S-T segment and T wave in rat electrocardiogram within 15 s ~ 5 min after injection of Pit, can significantly prolong the clotting time of rats. Conclusion LP patch has a significant protective effect on acute myocardial ischemia induced by Pit, and has the effect of inhibiting blood clotting and platelet aggregation.
