采用单克隆抗体(单抗)和WHO67/97标准血清建立ELISA法测定人血清免疫球蛋白G(IgG)亚类浓度。结果表明:本法敏感(最低可测值为2.1ng/ml,可测范围为100倍),精确(不同批测定平均变异系数≤8.06%)。所用单抗特异性强,反应性好,IgG亚类之和与常规单扩法所测总IgG值密切相关(r=0.96,P<0.01).强调将单抗包被于经BSA-戊二醛处理的微孔板上,能明显提高单抗吸附力和反应性;两种单抗混合包被可增加IgG亚类同种异型覆盖面。
Human serum immunoglobulin G (IgG) subclass concentrations were determined by ELISA using monoclonal antibodies (mAb) and WHO67 / 97 standard sera. The results showed that the method was sensitive (the lowest measurable value was 2.1ng / ml, the measurable range was 100 times), and the accuracy was accurate (the average coefficient of variation was ≤8.06% in different batches). The McAb specificity and reactivity were good, and the sum of IgG subclasses was closely related to the total IgG value measured by routine single-spike method (r = 0.96, P <0.01). Emphasized that the monoclonal antibody coated on BSA-glutaraldehyde-treated microplates, can significantly improve the mAb adsorption and reactivity; two monoclonal antibody hybrid coating can increase the IgG subclass allotype coverage.
