目的探讨Pin1和Her2在乳腺浸润性导管癌组织中的表达情况、二者之间的相关性及其临床意义,为乳腺肿瘤的防治提供参考。方法收集2012年3月到2015年5月在惠州市第一人民医院行外科手术切除的226例确诊的乳腺癌标本和50例正常乳腺组织,用免疫组织化学方法检测这些石蜡切片中Pin1和Her2蛋白表达,分析两者的相关性以及两者的表达与乳腺癌患者临床病理特征的关系。结果 Pin1蛋白和Her2蛋白在乳腺癌中表达的阳性率均远远高于正常乳腺(P<0.05)。Pin1的过表达与病理组织学分级、淋巴结有无转移相关,而与年龄、肿瘤大小、临床PTNM分期无明显相关性。Her2蛋白过表达与临床PTNM分期、淋巴结转移有相关性(P<0.05),与病理类型、年龄、肿瘤大小无明显相关性。在乳腺浸润性导管癌组织中Pin1和Her2蛋白阳性表达呈显著正相关(P<0.05)。结论 Pin1与Her2的过表达水平密切相关;Pin1和Her2在浸润性乳腺癌中发挥的重要作用,使Pin1可能成为Her2阳性的乳腺癌的一个新的治疗靶点。
Objective To investigate the expression of Pin1 and Her2 in infiltrating ductal carcinoma of the breast, the correlation between them and their clinical significance, to provide references for the prevention and treatment of breast tumors. Methods Totally 226 confirmed breast cancer specimens and 50 normal breast tissues were collected from March 2012 to May 2015 in Huizhou First People’s Hospital. Pin1 and Her2 in these paraffin sections were detected by immunohistochemistry Protein expression, analysis of the correlation between the two and the relationship between the two expression and clinicopathological features of breast cancer patients. Results The positive rates of Pin1 protein and Her2 protein expression in breast cancer were much higher than those in normal breast (P <0.05). Pin1 overexpression and histopathological grading, lymph node metastasis related, but with age, tumor size, clinical PTNM stage no significant correlation. The overexpression of Her2 protein was correlated with clinical stage, lymph node metastasis and PTNM (P <0.05), but not with pathological type, age and tumor size. The positive expression of Pin1 and Her2 in breast invasive ductal carcinoma was significantly and positively correlated (P <0.05). Conclusion Pin1 and Her2 overexpression are closely related to Pin1 and Her2 in invasive breast cancer play an important role in the Pin1 may be Her2-positive breast cancer a new therapeutic target.
