以大豆蚜(Aphis glycines)为材料,比较分析了氯仿-异戊醇抽提法、盐析法、KAc法及试剂盒法4种基因组DNA提取方法。结果表明:在提取大豆蚜基因组DNA的纯度上,氯仿-异戊醇抽提法和盐析法较高;提取所用时间由短到长依次为试剂盒提取法、盐析法、氯仿-异戊醇抽提法和KAc法。盐析法能够更快速有效地提取大豆蚜基因组DNA,是一种值得推广应用的基因组DNA提取方法,另外3种方法也有一定的优势,应根据试验条件和要求选择相应的基因组DNA提取方法。
Aphis glycines were used as materials to analyze four kinds of genomic DNA extraction methods, such as chloroform-isoamyl alcohol extraction method, salting-out method, KAc method and kit method. The results showed that chloroform-isoamyl alcohol extraction and salting-out were the best methods for extracting the genome DNA of soybean aphid. The extraction time was from short to long followed by kit extraction, salting-out, chloroform- Alcohol extraction method and KAc method. Salting-out method can extract genomic DNA of soybean aphid more quickly and efficiently. It is a genomic DNA extraction method worthy of popularization and application. The other three methods also have certain advantages. Corresponding genomic DNA extraction methods should be selected according to the test conditions and requirements.
