目的分析长沙地区汉族人群脑出血患者血脂水平与载脂蛋白B基因(apolipoprotein B,apoB)G12669A多态性的关系。方法收集93例散发性脑出血患者和100例正常对照者的外周血标本。采用氧化酶法测定血清甘油三酯(triglycerides,TG)、总胆固醇(total cholesterol,TC)、高密度脂蛋白(high density lipoprotein cholesterol,HDL)、低密度脂蛋白(low density lipoprotein cholesterol,LDL)水平,酶联免疫吸附法测定LP(a)浓度,免疫比浊法测定apoB100及apoAI浓度;聚合酶链式反应-限制性片段长度多态性分析法(Polymerase Chain Reaction-Restriction Fragment Length Polymorphism,PCR-RFLP)和DNA直接测序法检测apoB基因G12669A多态性。结果①长沙地区汉族人群脑出血患者的TC、TG、LDL-C和LP(a)显著高于正常对照组,而HDL-C显著降低(P<0.05);②脑出血组中,G/A基因型的TC、TG、LDL-C较之G/G型显著增高,而HDL-C则显著降低(P<0.05),其他指标无显著性差异(P>0.05);③脑出血组及对照组apoB基因G12669A多态A等位基因频率分别为0.097、0.045,脑出血组apoB基因G12669A多态A等位基因频率显著高于对照组(P<0.05)。结论长沙地区汉族人群脑出血患者的血脂水平升高,可能与apoB基因G12669A多态A等位基因存在相关性。
Objective To analyze the relationship between plasma lipid level and apolipoprotein B (apoB) G12669A polymorphism in patients with cerebral hemorrhage in Han nationality in Changsha area. Methods Peripheral blood samples from 93 patients with sporadic cerebral hemorrhage and 100 normal controls were collected. The levels of triglycerides (TG), total cholesterol (TC), high density lipoprotein cholesterol (HDL) and low density lipoprotein cholesterol (LDL) , The concentrations of apoB100 and apoAI were determined by enzyme-linked immunosorbent assay (ELISA), the concentrations of apoB100 and apoAI by immunoturbidimetric assay. The levels of apoB100 and apoAI were determined by polymerase chain reaction-restriction fragment length polymorphism (PCR- RFLP) and direct DNA sequencing to detect apoB gene G12669A polymorphism. Results ① The levels of TC, TG, LDL-C and LP (a) in patients with ICH in Changsha were significantly higher than those in controls and HDL-C (P <0.05) TC, TG and LDL-C of genotypes were significantly higher than those of G / G and HDL-C (P <0.05), but there was no significant difference between other indexes (P> 0.05) The frequency of polymorphism A allele of apoB gene G12669A was 0.097,0.045 in group A and B respectively. The frequency of the allele G12669A polymorphism in apoB gene group was significantly higher than that in control group (P <0.05). Conclusion The level of blood lipid in patients with cerebral hemorrhage in Han nationality in Changsha area is elevated, which may be related to the polymorphism A allele of apoB gene G12669A.
