紫杉烯合成酶(taxadiene synthase)是抗癌药物紫杉醇生物合成途径的第一个酶,也是关键酶,在红豆杉植物体内催化生成紫杉醇的母核——紫杉烯;它在植物体内低水平的表达和低酶活力被认为是紫杉醇在植物体内含量低微的重要因素。Wildung等已经得到了太平洋红豆杉(Taxus brevifolia)紫杉烯合成酶的cDNA及其核苷酸顺序,长度为2700bp,开放阅读框架为2586bp,其中包括编码137个氨基酸残基的质体转移肽和725个氨基酸残基的紫杉烯合成酶肽链。我们过去的研究得到一中国红豆杉(T.chinensis)高产紫杉烷类化合物的愈伤组织细胞系,并构建了它的cDNA文库,克隆载体为λgtlO。
