目的观察1,25二羟基维生素D3[1,25(OH)_2D_3]对HepG_2细胞内TG含量及其代谢途径中相关基因脂联素受体2(AdipoR2)、p38丝分裂素激活蛋白激酶(p38MAPK)、脂蛋白脂(LPL)表达的影响,揭示1,25(OH)_2D_3在调控HepG_2细胞内TG代谢中的作用,并探讨1,25(OH)_2D_3调控TG代谢的可能机制。方法将HepG_2细胞分为Con组、溶剂组、MK886组、VitD组及VitD+MK886组。检测各组肝细胞内AdipoR2、p38MAPK、LPL基因、蛋白表达水平及肝细胞内TG含量。结果 Con组、溶剂组、VitD+MK886组比较,3种目标基因及蛋白含量、TG含量差异均无统计学意义(P>0.05);MK886组与Con组、溶剂组、VitD+MK886组比较,3种目标基因及蛋白含量降低(P<0.05),而TG含量升高[(841.26±60.1)vs(432.17±58.23)vs(427.18±57.85)vs(429.36±57.96)mg/g,P<0.05];VitD组与Con组、溶剂组、VitD+MK886组比较,3种目的基因及蛋白含量升高(P<0.05),而TG含量降低[(231.72±40.83)vs(432.17±58.23)vs(427.18±57.85)vs(429.36±57.96)mg/g,P<0.05];VitD组与MK886组比较,3种目的基因及蛋白含量升高(P<0.05),而TG含量降低[(231.72±40.83)vs(841.26±60.13)mg/g,P<0.05]。结论 1,25(OH)_2D_3可能是通过上调AdipoR2、p38MAPK、LPL的表达来降低肝细胞内TG含量。
Objective To investigate the effect of 1,25 dihydroxy vitamin D3 [1,25 (OH) _2D_3] on TG content in HepG2 cells and its related genes adiponectin receptor 2 (AdipoR2), p38 mitogen-activated protein kinase ), Lipoprotein (LPL) expression, revealing the role of 1,25 (OH) _2D_3 in the regulation of TG metabolism in HepG_2 cells, and to explore the possible mechanism of 1,25 (OH) _2D_3 regulation of TG metabolism. Methods HepG2 cells were divided into Con group, solvent group, MK886 group, VitD group and VitD + MK886 group. The levels of AdipoR2, p38MAPK and LPL, the protein expression and the content of TG in liver cells of each group were detected. Results Compared with the Con group, the solvent group and the VitD + MK886 group, there was no significant difference in the three target gene, protein content and TG content among the Con group, solvent group and VitD + MK886 group (P> 0.05) (P <0.05), while the content of TG, TG and TG in three kinds of target genes decreased (P <0.05), and the content of TG increased (841.26 ± 60.1 vs 432.17 ± 58.23 vs 427.18 ± 57.85 vs 429.36 ± 57.96 mg / (P <0.05), but the content of TG decreased ([231.72 ± 40.83] vs (432.17 ± 58.23) vs (P <0.05), compared with that of Con group, solvent group and VitD + MK886 group 427.18 ± 57.85 vs 429.36 ± 57.96 mg / g, P <0.05]. Compared with the MK886 group, the levels of three genes and proteins in VitD group were significantly increased (P <0.05) ) vs (841.26 ± 60.13) mg / g, P <0.05]. Conclusion 1,25 (OH) _2D_3 may decrease the content of TG in hepatocytes by up-regulating the expression of AdipoR2, p38MAPK and LPL.
