采用活化的高效液相膜亲和介质为固相化载体,与纯化的人IgM 抗体进行交联,制备了IgM 高效膜亲和色谱柱,与IgG膜亲和柱比较了交联效率,并用相应的抗抗体通过HPLC测定了两者的亲和能力。在相同条件下,IgM 的交联效率为IgG的一半,但最大亲和容量却与之相近,并且在一定范围内可以完全分离血清中抗IgM 抗体。结果表明:IgM 高效膜色谱柱可以高效快速亲和分离相应抗体
An activated high-performance liquid phase membrane affinity medium is used as a solid phase carrier and is cross-linked with the purified human IgM antibody to prepare an IgM high-performance membrane affinity chromatography column. The efficiency of crosslinking is compared with that of an IgG membrane affinity column, The anti-antibodies were measured by HPLC affinity between the two.
