目的探讨CD4+CD25highT细胞(Treg)在60例上皮性卵巢癌患者外周血中的分布及其表型特征,分析手术对卵巢癌患者外周血Treg的影响。方法采用流式细胞术检测卵巢癌患者及健康人外周血中Treg的比例,探讨其表型特征并分析手术对患者外周血Treg比例的影响。结果与正常对照(2.95±1.09)%相比,60例上皮性卵巢癌患者外周血中Treg比例明显增加(9.81±5.19)%,二者差异有统计学意义(P<0.01);手术后,患者外周血中Treg的比例相比术前显著下降(P<0.01);Treg表型分析发现:Treg高表达CD45RO、HLA-Ⅰ、Foxp3分子,较高水平表达OX40、GITR、CD152、CD95、CD95L、B7-H1,几乎不表达CD80、CD86、B7-H4、CD45RA、CD69分子,而HLA-DR、CD154的表达水平在个体间差异较大,同时大部分Treg细胞CD127表达阴性。结论上皮性卵巢癌患者外周血CD4+CD25highT细胞水平明显升高,可能参与卵巢癌患者肿瘤免疫抑制。
Objective To investigate the distribution and phenotypic characteristics of CD4 + CD25highT cells (Tregs) in peripheral blood of 60 patients with epithelial ovarian cancer and analyze the effect of surgery on Tregs in peripheral blood of patients with ovarian cancer. Methods Flow cytometry was used to detect the proportion of Treg in peripheral blood of patients with ovarian cancer and healthy people. The phenotypic characteristics and the effect of surgery on the percentage of Treg in peripheral blood of patients with ovarian cancer were analyzed. Results Compared with the normal control (2.95 ± 1.09)%, the percentage of Treg in the peripheral blood of 60 patients with epithelial ovarian cancer was significantly increased (9.81 ± 5.19)%, the difference was statistically significant (P <0.01). After the operation, The percentage of Tregs in peripheral blood of patients was significantly lower than that before operation (P <0.01). Treg phenotype analysis showed that Tregs overexpressed CD45RO, HLA-Ⅰ and Foxp3 and expressed OX40, GITR, CD152, CD95 and CD95L , B7-H1, almost no expression of CD80, CD86, B7-H4, CD45RA, CD69 molecules, and HLA-DR, CD154 expression levels vary between individuals, while most of Treg cells CD127 negative expression. Conclusion The level of CD4 + CD25highT cells in peripheral blood of patients with epithelial ovarian cancer is significantly increased, which may be involved in tumor immunosuppression in patients with ovarian cancer.
