目的探讨脊髓神经元自噬与大鼠糖尿病神经病理性疼痛(DNP)的关系。方法选取成年雄性健康SPF级SD大鼠60只,随机分为造模组50只和对照组10只。比较两组不同时间点血糖、机械疼痛阈值、LC3/TUNEL阳性神经元细胞数、LC3-Ⅱ蛋白表达和LC3-Ⅱ/LC3-Ⅰ比值的差异,以及各指标间的相关性。结果造模后,造模组血糖值随时间而升高,且高于对照组(P<0.05),机械阈值及热刺疼痛阈值均随时间而降低,且低于对照组(P<0.05);LC3/TUNEL阳性神经元细胞数,LC3-Ⅱ蛋白表达和LC3-Ⅱ/LC3-Ⅰ比值随时间而升高,且在不同时间点均高于对照组(P<0.05)。相关分析显示,血糖与LC3/TUNEL阳性神经元细胞数、LC3-Ⅱ蛋白表达、LC3-Ⅱ/LC3-Ⅰ比值呈正相关(P<0.05),机械疼痛阈值与LC3/TUNEL阳性神经元细胞数、LC3-Ⅱ蛋白表达、LC3-Ⅱ/LC3-Ⅰ比值呈负相关(P<0.05)。结论大鼠DNP模型中脊髓神经元自噬功能增强,提示其可能与DNH有关。
Objective To investigate the relationship between spinal cord autophagy and diabetic neuropathic pain (DNP) in rats. Methods Sixty male Sprague-Dawley (SD) SD rats were randomly divided into model group (n = 50) and control group (n = 10). The differences of blood glucose, mechanical pain threshold, LC3 / TUNEL positive neurons, LC3-Ⅱ protein expression and LC3-Ⅱ / LC3-Ⅰ ratio at different time points were compared between the two groups. Results After modeling, blood glucose of model group increased with time and was higher than that of control group (P <0.05). The mechanical threshold and the pain threshold of Tottenham were all decreased with time and were lower than those of control group (P <0.05) ; The ratio of LC3 / TUNEL positive neurons, the expression of LC3-Ⅱ and the ratio of LC3-Ⅱ / LC3-Ⅰ increased with time, and were higher than those in the control group at different time points (P <0.05). Correlation analysis showed that there was a positive correlation between blood glucose and the number of LC3 / TUNEL positive neurons, the expression of LC3-Ⅱ protein and the ratio of LC3-Ⅱ / LC3-Ⅰ (P <0.05) .The mechanical pain threshold was positively correlated with the number of LC3 / TUNEL positive neurons, LC3-Ⅱ protein expression, LC3-Ⅱ / LC3-Ⅰ ratio was negatively correlated (P <0.05). Conclusion The autonomic function of spinal cord neurons is enhanced in rat DNP model, suggesting that it may be related to DNH.
