采用分光光度法,以邻硝基苯基荧光酮(O-NPF)为显色剂,研究了O-NPF与头孢唑啉(CFZ)络合物的可见光谱性质及测定条件。结果表明:CFZ在4.545~72.72μg/mL范围内遵循比尔定律,回归方程为:A=0.668ρ+0.5341,检出限1.53μg/mL,表观摩尔吸光系数为5.41×104L.mol-1.cm-1,相关系数为R2=0.9891,并探讨了其反应机理。方法已用于尿样及生物样品中CFZ含量的测定。
The spectroscopic properties and determination conditions of O-NPF and cefazolin (CFZ) complexes were studied by using spectrophotometry with o-nitrophenylfluorone (O-NPF) as chromogenic reagent. The results showed that the CFZ obeys Beer’s law in the range of 4.545 ~ 72.72μg / mL. The regression equation was: A = 0.668ρ + 0.5341, the detection limit was 1.53μg / mL and the apparent molar absorptivity was 5.41 × 104L.mol-1. cm-1, the correlation coefficient is R2 = 0.9891, and the reaction mechanism was discussed. The method has been applied to the determination of CFZ in urine samples and biological samples.
