目的研究bcl-2反义寡核苷酸在抑制HL-60细胞增殖和下调细胞内bcl-2蛋白表达水平的作用。方法将人工合成的bcl-2反义寡核苷酸片段与HL-60细胞共培养,在作用的第0天、1天、3天、5天通过细胞计数和锥虫蓝染色法检测细胞的生长、存活情况,同时通过免疫组化法(APAAP法)检测细胞内bcl-2蛋白表达水平的变化情况。结果bcl-2正、反义寡核苷酸均可抑制HL-60细胞增殖,但bcl-2反义寡核苷酸的作用更为显著。bcl-2反义寡核苷酸可下调HL-60细胞内bcl-2的蛋白表达水平,其作用与其浓度和时间呈正相关,而bcl-2正义寡核苷酸及对照无此作用。结论bcl-2反义寡核苷酸可以有效地抑制HL-60细胞的增殖和降低细胞内bcl-2的蛋白表达水平。
Objective To investigate the effect of bcl-2 antisense oligodeoxynucleotides on inhibiting the proliferation of HL-60 cells and down-regulating the expression of bcl-2 protein in cells. Methods Synthesized bcl-2 antisense oligonucleotides were co-cultured with HL-60 cells. The cells were detected by cell counting and trypan blue staining on the 0th, 1st, 3rd, and 5th day of action. Growth and survival conditions were also tested by immunohistochemical method (APAAP method) to detect changes in the expression of bcl-2 protein in cells. Results Both bcl-2 positive and antisense oligonucleotides could inhibit the proliferation of HL-60 cells, but the effect of bcl-2 antisense oligonucleotides was more significant. The bcl-2 antisense oligonucleotide can down-regulate the expression of bcl-2 protein in HL-60 cells, and its effect is positively correlated with its concentration and time. However, bcl-2 sense oligonucleotide and control have no such effect. Conclusion bcl-2 antisense oligonucleotide can effectively inhibit the proliferation of HL-60 cells and reduce the expression of bcl-2 protein in cells.
