研究肿瘤坏死因子受体相关因子(TRAF)在原发性胆汁性肝硬化(PBC)患者外周血单个核细胞(PBMC)中的表达及其与疾病发生发展的关系。分别采用实时荧光定量PCR和免疫印迹法,检测47例PBC和47例健康对照者中TRAF1和TRAF2的基因和蛋白表达。结果表明:PBC患者PBMC中TRAF2 mRNA和蛋白的表达明显高于健康对照组(P<0.05),而TRAF1在两者中表达无明显区别。PBC患者Ⅲ、Ⅳ期PBMC中的TRAF2的mRNA和蛋白的表达较Ⅰ、Ⅱ期明显增高(P<0.05);提示:TRAF2的表达与PBC的发生发展存在一定的相关性,在PBC的发病机制起到重要的作用,为PBC的诊断和预防提供了新线索。
To investigate the expression of tumor necrosis factor receptor related factor (TRAF) in peripheral blood mononuclear cells (PBMCs) of patients with primary biliary cirrhosis (PBC) and its relationship with the occurrence and development of the disease. Real-time fluorescence quantitative PCR and Western blotting were used to detect the gene and protein expression of TRAF1 and TRAF2 in 47 cases of PBC and 47 cases of healthy controls. The results showed that the expression of TRAF2 mRNA and protein in PBMC of patients with PBC was significantly higher than that of healthy controls (P <0.05), while TRAF1 expression was not significantly different between the two groups. The mRNA and protein expression of TRAF2 in PBMCs of stage Ⅲ and Ⅳ in PBC patients were significantly higher than those in stage Ⅰ and Ⅱ (P <0.05), which indicated that the expression of TRAF2 correlated with the occurrence and development of PBC. Play an important role in the diagnosis and prevention of PBC provides a new clue.
