目的研究黄芪对创伤性脑损伤后脑EB含量和紧密连接相关蛋白claudin-5表达的影响。方法采用自由落体致伤原理制成大鼠脑损伤模型,伊文思蓝法检测BBB通透性,Western blot法检测Claudin-5的表达。结果脑损伤模型组及黄芪组BBB通透性于脑损伤后明显增加,同时黄芪组BBB通透性低于脑损伤模型组;Claudin-5的表达在脑损伤模型组和黄芪组于伤后明显减少,黄芪组的表达明显高于脑损伤模型组(P<0.01)。结论调节紧密连接相关蛋白Claudin-5的表达,进而调节BBB的通透性可能是黄芪抑制脑水肿形成的途径之一。
Objective To study the effect of Astragalus on the content of EB and the expression of claudin-5 in the brain following traumatic brain injury. Methods The rat model of brain injury was established by free-fall injury. The permeability of BBB was detected by Evans blue method. The expression of Claudin-5 was detected by Western blot. Results The permeability of BBB in brain injury model group and astragalus group increased significantly after brain injury, meanwhile BBB permeability in Astragalus group was lower than that in brain injury model group. The expression of Claudin-5 in brain injury model group and Astragalus group was significantly The expression of Astragalus membranaceus was significantly higher than that of the brain injury model group (P <0.01). Conclusions The regulation of Claudin-5 expression and the regulation of BBB permeability may be one of the ways to inhibit the formation of cerebral edema.
