目的通过检测汉滩病毒(HTNV)76-118株感染SCID小鼠组织中的特异性病毒抗原、核酸的分布以及病理学变化,以建立汉滩病毒感染动物的评价体系。方法取汉滩病毒76-118株病毒液通过肌肉注射接种SCID小鼠,接种3 d后,取小鼠的心、肝、脾、肺、肾、脑等组织,并分别利用ELISA法检测各组织中的病毒特异性抗原;利用q RT-PCR法检测各组织中的病毒RNA;利用HE染色法检测各组织中的病理学变化。结果SCID小鼠感染汉滩病毒后,用夹心ELISA法可在其心、肝、脾、肺、肾组织中检测到HTNV特异性抗原,平均P/N值分别为2.33、7.41、8.13、7.40、2.89,其中脾病毒抗原含量最高;以q RT-PCR法检测小鼠各脏器组织中的HTNV核酸,在实验组小鼠的心、肝、脾、肺、肾中均可检测到病毒核酸,与对照组相比,其相对量分别达到2~(2.11)、2~(6.23)、2~(8.21)、2~(5.26)、2~(3.79)倍,其中脾病毒核酸含量最多;HE染色法可见肝、脾、肺中伴有显著的病理学变化。结论成功检测了汉滩病毒感染SCID小鼠后组织中特异性抗原、核酸的分布以及病理学变化,从而为建立汉滩病毒感染动物的评价体系提供一种参考。
Objective To detect the specific virus antigens, nucleic acid distribution and pathological changes in SCID mice infected by HTNV strain 76-118 to establish the evaluation system of Hantaan virus-infected animals. Methods SCID mice were inoculated intramuscularly with Hantaan virus strain 76-118. After 3 days of inoculation, the tissues of heart, liver, spleen, lung, kidney and brain of mice were inoculated. Results After SCID mice were infected with Hantaan virus, HTNV specific antigens were detected by sandwich ELISA in hearts, liver, spleen, lung and kidney. The average P / N values were 2.33, 7.41, 8.13, 7.40, 2.89, of which the spleen virus antigen content was the highest. The HTNV nucleic acids in various tissues of mice were detected by q RT-PCR.
