我院于1979年开始根据国内外文献所提供的方法,改用超过滤法,先后研制成牛、猪胸腺素组分V(THF_5),经冻干制成粉针剂。几年来,经动物实验及临床观察,发现对抗癌、抗放、抗衰老等有一定临床作用。现将研究结果报告如下:一、THF_5生产工艺及质量检测我院根据Goldstein Hooper及刘士廉等的方法,将牛、猪胸腺经抽提、盐析、脱盐、超滤、分装制成冻干THF_5粉针剂,经各项试验证实有活性后方供临床使用。二、THF_5动物试验情况1.猪、牛THF_5生物特性对比为观察猪、牛胸腺素特性,经分析发现两者蛋白质含量基本一致,对活性玫瑰花结的影响,猪THF_5活性略低于牛THF_5。两者对骨髓中T细胞分化的影响无甚差别,两种制剂经SDS电泳与聚焦电泳,发现它们分子量基本一致。2.THF_5对造血系统的作用(1)THF_5对小鼠粒系造血祖细胞的增殖作用选择24~28克雄性瑞士纯种小鼠,每组6只,1~5组,每只小鼠分别用THF_5 5μg连续腹腔注射5、10、15、20、25天,第6组不处理作为对照组,两组选择条件相同。
