目的:观察解毒消癥饮含药血清抑制胃腺癌细胞增殖的相关因素。方法:采用血清药理学方法,MTT法检测含药血清对人胃腺癌细胞的增殖影响,采用化学比色法、ELISA、R IA检测解毒消癥饮含药血清的NO、NOS、TNF-α、IL-1β、VEGF和内毒素含量。结果:MTT实验结果显示,与胎牛血清对照组比较,含药血清组可以显著抑制胃腺癌细胞的生长,且呈时间剂量依赖;各组血清的内毒素水平均符合胎牛血清的质量标准;与空白组比较,NO、NOS、TNF-α、IL-1β有显著差异,各组VEGF的水平没有差异;而且NO、NOS差异与胃腺癌细胞增殖抑制率具有较高的相关性。结论:解毒消癥饮含药血清的NO、NOS水平与其药理作用密切相关,可作为其含药血清的质量标准之一。
Objective: To observe the related factors of inhibiting the proliferation of gastric adenocarcinoma cells with Jiedu Xiaozheng decoction serum. Methods: Serum pharmacological methods and MTT assay were used to detect the effects of serum containing drugs on the proliferation of human gastric adenocarcinoma cells. The contents of NO, NOS, TNF-α, IL-1β, VEGF and endotoxin content. Results: The results of MTT assay showed that compared with the control group, the drug-containing serum group could significantly inhibit the growth of gastric adenocarcinoma cells in a time and dose-dependent manner. The levels of endotoxin in serum of each group were in accordance with the quality standards of fetal bovine serum. Compared with the blank group, NO, NOS, TNF-α, IL-1β were significantly different, and there was no difference in the levels of VEGF between the groups. Moreover, the differences of NO and NOS were highly correlated with the proliferation inhibition rate of gastric adenocarcinoma cells. Conclusion: The levels of NO and NOS in Jiedu Xiaozheng Decoction containing serum are closely related to its pharmacological effects, which can be used as one of the quality standards of its containing serum.
