目的:建立同时测定雷公藤类药材中雷公藤甲素、雷酚内酯、去甲泽拉木醛、雷公藤红素的HPLC-PDA分析方法。方法:采用乙酸乙酯超声法提取雷公藤类药材中的萜类成分;采用HPLC-PDA法对其进行分析测定,色谱柱:Dimonsiol C18柱(250 mm×4.6 mm,5μm);柱温25℃;流速:1.0 m L/min;以流动相乙腈-0.1%甲酸水进行梯度洗脱。结果:4种萜类成分在各自浓度范围内线性关系良好(0.9994≤r≤0.9999),平均加样回收率为92.17%~101.32%,RSD为1.37%~3.78%。结论:建立的HPLC-PDA同时分析4种萜类成分的方法灵敏度高、准确度可靠,适用于雷公藤类药材中萜类成分的分析测定。
Objective: To establish a HPLC-PDA method for simultaneous determination of triptolide, triptolide, desmethylselaldehyde and tripterine in Tripterygium wilfordii. Methods: The terpenoids in Tripterygium wilfordii were extracted by ethyl acetate. The HPLC-PDA method was used to determine the terpenoids. The column was Dimonsiol C18 (250 mm × 4.6 mm, 5 μm). The column temperature was 25 ℃ ; Flow rate: 1.0 m L / min; gradient elution with mobile phase of acetonitrile-0.1% formic acid in water. Results: The four terpenoids showed good linearity (0.9994≤r≤0.9999) in the range of their concentration. The average recoveries were 92.17% ~ 101.32% and the RSDs were 1.37% ~ 3.78%. Conclusion: The established HPLC-PDA method for simultaneous analysis of four terpenoids has high sensitivity and accuracy, and is suitable for the determination of terpenoids in Tripterygium wilfordii.
