目的:研究Ada蛋白质对alkA基因表达的调节,探讨Ada蛋白质与alkA基因有效转录的关系。方法:采用DNA顺序测定和体外转录的方法,以lac基因启动子为对照进行实验。结果:Ada蛋白质加入以突变和野生alkA基因启动子为模板的体外转录体系中,产生的alkAmRNA高于无Ada蛋白质组;Ada蛋白质加入lac基因启动子转录体系中,产生的lacmRNA与未加Ada蛋白质组基本相同。结论:Ada蛋白质对lac基因启动子转录调节作用不明显,对于突变的和野生的alkA基因转录均有促进作用,这种调节作用可能发生在启动子上游区。
AIM: To investigate the regulation of alka gene expression by Ada protein and to explore the relationship between Ada protein and alkA gene transcription. Methods: DNA sequence determination and in vitro transcription method, lac gene promoter as a control experiment. Results: Ada protein was added into the in vitro transcription system with mutant and wild-type alkA promoter as template. The resulting alkA mRNA was higher than that without Ada protein. When Ada protein was added into the lac gene promoter transcription system, laccRNA produced was significantly higher than that without Ada protein The group is basically the same. CONCLUSION: The Ada protein has no obvious effect on the transcriptional regulation of lac gene promoter, which can promote the transcription of mutant and wild alkA gene. This regulation may take place in the promoter upstream region.
