对荧光涂料激发能源147Pm辐照人淋巴细胞白血病细胞株Molt-4细胞和巨噬细胞株Ana-1细胞诱发的细胞凋亡进行了研究。实验中估算了147Pm在不同阶段培养细胞中的辐射累积吸收剂量。通过透射电镜的形态观察,发现Molt-4和Ana-1两株免疫细胞在受147Pm辐照时,可诱发明显的核断裂,核染质边聚,以及呈现膜包裹着的凋亡小体形成为特征的细胞凋亡。进而抽提了Molt-4和Ana-1两株免疫细胞的DNA,进行琼脂糖凝胶电泳也显示出阶梯状条带形成的细胞凋亡特征。并且观察到147Pm诱发的免疫细胞凋亡与147Pm辐照的时间和累积吸收剂量相关。
Apoptosis induced by fluorescent coating excitation energy 147Pm irradiated human lymphoblastic leukemia cell line Molt-4 and macrophage cell line Ana-1 were studied. In the experiment 147Pm estimated in different stages of cultured cells in radiation absorbed absorbed dose. Morphological observation by transmission electron microscopy revealed that Molt-4 and Ana-1 immune cells could induce obvious nuclear breaks, nuclear edge-aggregates, and apoptotic bodies that were surrounded by membrane-bound apoptotic bodies when exposed to 147 Pm Characteristics of apoptosis. Furthermore, the DNA of Molt-4 and Ana-1 immune cells was extracted. The agarose gel electrophoresis also showed the apoptotic characteristics of the ladder-shaped bands. And it was observed that 147 Pm-induced apoptosis of immune cells correlated with the time of 147 Pm irradiation and the cumulative absorbed dose.
