本文报道用最小浓度的乙胺嘧啶抑制体外培养的恶性疟原虫无性体增殖,从而获得纯配子体的方法。同时将此培养纯配子体的方法用于观察Ⅲ期配子体对氯喹、卤泛曲林、乙胺嘧啶、奎宁的敏感性。使用的恶性疟原虫为NF54株,它对氯喹、乙胺嘧啶、卤泛曲林、奎宁均敏感。以不同方法配制药液。用蒸馏水配制1×10~(-3)mol/L的氯喹,0.5%乳酸配制1×10~(-2)mol/L乙胺嘧啶,70%乙醇配制2×10~(-3)mol/L卤泛曲林,蒸馏水配制1×10~(-3)mol/L的奎宁。用RPMI 1640完全培养液按不同浓度稀释药液。
In this paper, we reported that pyrimethamine with the minimum concentration inhibits the growth of P. falciparum asexual cells cultured in vitro to obtain a pure gametophyte. At the same time, the method of cultivating pure gametophyte was used to observe the sensitivity of stage Ⅲ gametophyte to chloroquine, halibutine, pyrimethamine and quinine. Plasmodium falciparum used was NF54 strain, which is sensitive to chloroquine, pyrimethamine, halibutine and quinine. Prepare the solution in different ways.
