目的:探讨穿心莲煎剂干预治疗变应性鼻炎的效果及可能作用机制。方法:建立豚鼠变应性鼻炎模型,实验设空白组、模型组,穿心莲组、西替利嗪组和联合用药组,并给予相应药物灌胃干预。运用HE染色、ELISA、RT-PCR法分别观察各组动物鼻腔黏膜和肺组织病理学变化、血清中细胞因子的含量及肺组织中Tim1、Tim3的mRNA表达。结果:穿心莲煎剂、西替利嗪和两者联合用药均能不同程度降低变应性鼻炎豚鼠症状学积分;改善鼻腔黏膜、肺组织、肺泡灌洗液的炎症程度;上调血清中INF-γ含量,下调IL-4含量;下调肺组织中Tim1、Tim3的mRNA表达,且联合用药组效果最为显著。结论:穿心莲煎剂能够通过干预Th1/Th2细胞亚群平衡,进而改善鼻腔黏膜和肺组织的变应性炎症,且与西替利嗪联合用药效果更为显著。
Objective: To investigate the effect of Andrographis paniculata (Willow) decoction on the treatment of allergic rhinitis and its possible mechanism. Methods: The model of allergic rhinitis in guinea pigs was established. The experiment was divided into blank group, model group, andrographolide group, cetirizine group and combined treatment group, and given the corresponding drug gavage intervention. Pathological changes of nasal mucosa and lung tissue, contents of cytokines in serum and mRNA expressions of Tim1 and Tim3 in lung tissues were observed by HE staining, ELISA and RT-PCR respectively. Results: The combination of both andrographolide and cetirizine could reduce the symptom scores of allergic rhinitis guinea pigs to varying degrees; improve the inflammatory degree of nasal mucosa, lung tissue and bronchoalveolar lavage fluid; increase the serum INF-γ Content, down-regulate IL-4 content; down-regulate the expression of Tim1, Tim3 mRNA in lung tissue, and the combined effect is the most obvious. Conclusion: Andrographis paniculata can improve the allergic inflammation of nasal mucosa and lung tissue through the intervention of Th1 / Th2 cell subpopulation, and the combination therapy with cetirizine is more effective.
