研究转化生长因子α(TGFα)、表皮生长因子受体 (EGFR)反义寡聚核苷酸 (ASODN)对人大肠癌生长增殖的作用。反义TGFα、反义EGFR及无关对照ODN经阳离子脂质体包裹后转染人大肠癌HR8348细胞。采用噻唑蓝比色法 (MTT法 ) ,细胞周期分析 ,裸鼠体内接种等方法测定瘤细胞体内外增殖的变化。与对照组HR8348细胞相比 ,经TGFα反义ODN处理的HR8348细胞体外增殖速度减慢 (抑制率达 80 % )、DNA合成受抑、裸鼠体内成瘤率下降 (2 5 %∶ 10 0 % )。经EGFR反义ODN处理的HR8348细胞也得到了类似的结果。TGFα反义ODN、EGFR反义ODN均能有效抑制大肠癌细胞的体内外生长增殖。
To study the effect of transforming growth factor α (TGFα) and epidermal growth factor receptor (EGFR) antisense oligonucleotide (ASODN) on the proliferation and proliferation of human colorectal cancer. Antisense TGFα, antisense EGFR and unrelated control ODN were transfected into human colon cancer HR8348 cells by cationic liposomes. The proliferation of tumor cells in vitro and in vivo was measured by MTT method, cell cycle analysis and inoculation in nude mice. Compared with HR8348 cells in control group, the proliferation of HR8348 cells treated with TGFα antisense ODN slowed down in vitro (80% inhibition rate), DNA synthesis was inhibited, and the rate of tumorigenesis in nude mice decreased (25%: 100% ). HR8348 cells treated with EGFR antisense ODN also yielded similar results. Both TGFα antisense ODN and EGFR antisense ODN can effectively inhibit the growth and proliferation of colorectal cancer cells in vitro and in vivo.
