目的研究hS100A6(human S100A6)对人卵巢癌细胞系(低转移系HO8910和高转移系HO8910 PM)增殖、凋亡和Wnt/β-catenin信号途径的影响。方法通过MTT以及台盼蓝染色检测细胞增殖的变化;Hoechst检测细胞凋亡的变化;提取细胞总蛋白及核蛋白Western blot检测hS100A6对β-catenin的影响。结果 MTT及台盼蓝染色结果显示hS100A6抑制HO8910细胞和HO8910 PM细胞的增殖(P<0.05),Hoechst染色结果显示hS100A6同时促进这两系细胞的凋亡(P<0.05);Western blot显示hS100A6增加β-catenin的水平并促进其发生核转位。结论 hS100A6能抑制人卵巢癌细胞系HO8910和HO8910 PM增殖,促进其凋亡,同时激活Wnt/β-catenin信号途径。
Objective To investigate the effects of human S100A6 on the proliferation, apoptosis and Wnt / β-catenin signaling in human ovarian cancer cell lines HO8910 and HO8910 PM with low metastatic potential. Methods MTT and trypan blue staining were used to detect the cell proliferation. Hoechst assay was used to detect the changes of apoptosis. The total cellular protein and nuclear protein were extracted and Western blot was used to detect the effect of hS100A6 on β-catenin. Results The results of MTT assay and trypan blue staining showed that hS100A6 inhibited the proliferation of HO8910 and HO8910 PM cells (P <0.05). Hoechst staining showed that hS100A6 promoted the apoptosis of both cells (P <0.05). Western blot showed that hS100A6 increased β-catenin and promote its nuclear translocation. Conclusion hS100A6 can inhibit the proliferation and promote the apoptosis of human ovarian cancer cell lines HO8910 and HO8910 PM and activate the Wnt / β-catenin signaling pathway.
