目的:评价匹维溴胺对束缚应激大鼠离体结肠平滑肌条的影响并探讨其对各型IBS可能的作用机理。方法:成年SD大鼠随机分为模型组和对照组,模型组用自制硬塑胶模具束缚2 h。分别制作两组大鼠近端及远端结肠的环行肌和纵行肌条共64条(每组肌条32个,各肌层8个)。采用离体平滑肌条器官浴槽实验方法,观察不同浓度匹维溴胺对平滑肌条自发性收缩活动的影响。结果:匹维溴胺对模型组和对照组离体结肠平滑肌条自发性收缩活动均表现为抑制作用,但对模型组效应较对照组显著(近端结肠28.54±4.82vs7.48±1.65,21.75±1.00vs12.56±3.15;远端结肠15.71±5.27vs3.89±1.16,20.16±3.16vs7.56±1.96)(P<0.05)。匹维溴胺对近端结肠平滑肌收缩的抑制作用较远端结肠显著(P<0.05)。匹维溴胺对相同部位结肠环行肌与纵行肌的抑制作用无显著性差异(P>0.05)。结论:匹维溴胺能抑制束缚应激大鼠离体结肠平滑肌的收缩,且对近端结肠收缩的抑制作用显著高于远端结肠;但在相同部位,对环行肌和纵行肌收缩的抑制作用无显著性差异。
OBJECTIVE: To evaluate the effect of piroxicam on isolated smooth muscle strips of rats with restraint stress and to explore the possible mechanism of action of piroxicam on various types of IBS. Methods: Adult SD rats were randomly divided into model group and control group. The model group was restrained for 2 hours with homemade hard plastic mold. Sixty-four mesangial and longitudinal strips (32 in each group and 8 in each muscle) were made in the proximal and distal colon, respectively. The effects of different concentrations of Pivalivirrin on spontaneous contractile activity of smooth muscle strips were observed with the experimental method of organ baths in vitro. RESULTS: Pivoxil had inhibitory effects on spontaneous contractions of smooth muscle strips in vitro and in vivo in model and control groups, but the effect was more significant in model group than in control group (28.54 ± 4.82 vs 7.48 ± 1.65 and 21.75 in proximal colon) ± 1.00 vs 12.56 ± 3.15; distal colon 15.71 ± 5.27 vs 3.89 ± 1.16, 20.16 ± 3.16 vs 7.56 ± 1.96) (P <0.05). Pivalivirrhine inhibited the proximal colon smooth muscle contraction more significantly than the distal colon (P <0.05). Pivivirin had no significant difference in the inhibitory effect on the circumcircle and longitudinal muscle of the colon at the same site (P> 0.05). CONCLUSION: Pivobromine can inhibit the contractile contraction of isolated rat colon smooth muscle contraction, and the inhibition of proximal colon contraction was significantly higher than the distal colon; but in the same site, the ring muscle and longitudinal muscle contraction No significant difference in inhibition.
