目的探讨葛根素对HepG2细胞胰岛素抵抗的改善作用。方法用MTT法检测葛根素对细胞增殖的影响;将HepG2细胞置于10μg/ml胰岛素培养液中24 h复制胰岛素抵抗模型,用葡萄糖氧化酶法检测葛根素对胰岛素抵抗HepG2细胞葡萄糖消耗的影响;采用蒽酮法检测细胞内糖原含量。结果葛根素对HepG2细胞增殖无显著影响,并可促进胰岛素抵抗HepG2细胞的葡萄糖消耗,增加肝细胞糖元含量。结论葛根素可调节HepG2肝细胞糖代谢,明显改善肝细胞胰岛素抵抗。
Objective To investigate the effect of puerarin on insulin resistance in HepG2 cells. Methods The effect of puerarin on the proliferation of HepG2 cells was detected by MTT assay. The model of insulin resistance was induced by HepG2 cells in 10μg / ml insulin medium for 24 hours. The effect of puerarin on glucose depletion in HepG2 cells was detected by glucose oxidase method. Anthrone method was used to detect intracellular glycogen content. Results Puerarin had no significant effect on the proliferation of HepG2 cells, and promoted the insulin resistance of HepG2 cells and increased the content of glycogen in HepG2 cells. Conclusion Puerarin can regulate HepG2 hepatocyte glucose metabolism and significantly improve hepatocyte insulin resistance.
