利用地高辛标记CRNA探针原位杂交技术检测内毒素性急性肾损伤大鼠肾组织内皮细胞白细胞粘附因子-1(ELAM-1)mRNA的表达.结果发现;受检肾组织毛细血管网及小静脉血管内皮细胞呈现ELAM-1mRNA的广泛表达,其时程变化为给予内毒素后1h已出现弱表达,3~6h达高峰,24h内衰城近无.这种表达提示了内毒素性急性肾损伤早期肾小球毛细血管内大量中性粒细胞(PMN)滞留聚集的重要分子基础,对于解释中毒性休克时急性肾功能衰竭的发生机理有重要价值.
The expression of leukocyte adhesion molecule-1 (ELAM-1) mRNA in renal tissue of endotoxin-induced acute renal injury rats was detected by digoxigenin labeled CRNA probe in situ hybridization. The results showed that the capillary network of the kidneys and small venules endothelial cells showed extensive expression of ELAM-1 mRNA, the time course of 1h after the endotoxin has been weakly expressed, reached a peak at 3 ~ 6h, within 24 hours of decay no. This expression suggests an important molecular basis for the retention and accumulation of a large number of neutrophils (PMNs) in glomerular capillaries during endotoxic acute renal injury, which is of great value in explaining the pathogenesis of acute renal failure in toxic shock.
