目的:以腺病毒为载体,研究外源性突变型p27(p27mt)基因对大肠癌的体内抑制作用。方法:应用细胞移植法把人大肠癌SW480细胞接种于BALB/c裸鼠皮下,构建大肠癌裸鼠模型,将已成功构建的腺病毒介导的突变型p27(Ad-p27mt)及Lac-Z采用瘤体内直接注射法注射到瘤体内,与空白对照组比较,绘制肿瘤生长曲线,计算肿瘤生长抑制率,并进行细胞凋亡的相关检测。结果:Ad-p27mt基因治疗组肿瘤生长明显受到抑制,有更强的促凋亡作用,与Lac-Z组、空白对照组比较,差异有显著性(P<0.01)。结论:Ad-p27mt基因对大肠癌裸鼠模型的体内肿瘤有明显抑制作用。
Objective: To study the inhibitory effect of exogenous mutant p27 (p27mt) gene on colorectal cancer in adenovirus vector. Methods: Human colorectal cancer SW480 cells were inoculated subcutaneously in BALB / c nude mice by cell transplantation to construct a model of colorectal cancer in nude mice. The adenovirus-mediated mutant p27 (Ad-p27mt) and Lac-Z The tumor in vivo was injected by injection directly into the tumor. Compared with the blank control group, the tumor growth curve was drawn, the growth inhibition rate of the tumor was calculated, and the related detection of apoptosis was carried out. Results: Ad-p27mt gene therapy group was significantly inhibited tumor growth, and have a stronger role in promoting apoptosis, Lac-Z group and the blank control group, the difference was significant (P <0.01). Conclusion: The Ad-p27mt gene can significantly inhibit the in vivo tumor in nude mice model of colorectal cancer.
