角膜新生血管形成(CNV)是角膜移植术后发生排斥反应致手术失败的主要原因。我们研究了基因重组人白细胞介素2(IL-2)EVA-40缓释膜片植入家兔角膜诱发 CNV 的特性与组织病理学改变以及内源性过氧化物酶——二氨基联苯胺(DAB)细胞化学法显示 CNV。结果表明:IL-2能剂量依赖性地诱导 CNV 生长,500U IL-2缓释能在100%家兔眼诱导规律的 CNV 形成,IL-2诱导的CNV 与各种炎细胞特别是淋巴细胞密切相关,是一个不断变化的动态过程,炎症消失 CNV 逐渐退行。该方法易于定量、重复,是一较理想的免疫源性 CNV 模型。DAB 法可清晰地显示 CNV 的萌发,生长及退行形态。
Corneal neovascularization (CNV) is a major cause of failure of surgery after corneal transplant rejection. We investigated the characteristics and histopathological changes of CNV induced by corneal implantation of recombinant human interleukin-2 (IL-2) sustained-release membrane in rabbit cornea and the effects of endogenous peroxidase-diaminobenzidine (DAB) Cytochemistry shows CNV. The results showed that IL-2 could induce CNV growth in a dose-dependent manner, and sustained release of 500U IL-2 could induce regular CNV formation in 100% rabbit eyes. IL-2-induced CNV was closely associated with various inflammatory cells, especially lymphocytes Related, is a dynamic process of change, CNV gradually disappear after inflammation disappears. The method is easy to quantify and repeat, and is an ideal immunogenic CNV model. DAB method can clearly show the CNV germination, growth and degenerative morphology.
